We use DataFrame.copy() for copying a DataFrame instead of simply using assignment operator i.e, train_copy = strat_train_set for copying it. Why?

We can use either of the techniques. It doesn't matter.
Setting a variable using the assignment operator actually sets a reference to the DataFrame instead of copying it. So any changes made in the variable will reflect in the DataFrame.
We just want to clone the DataFrame(set a reference), instead of copying it. Cloning is achieved by DataFrame.copy() method.
DataFrame.copy() returns a DataFrame object while using assignment operator we get a python list.
